    On the Recovery and Resolution of Exponential Relaxation Rates from Experimental Data: A Singular-Value Analysis of the Laplace Transform Inversion in the Presence of Noise

    The problem of numerical inversion of the Laplace transform is considered when the inverse function is of bounded, strictly positive support. The recent eigenvalue analysis of McWhirter and Pike for infinite support has been generalized by numerical calculations of singular values. A priori knowledge of the support is shown to lead to increased resolution in the inversion, and the number of exponentials that can be recovered in given levels of noise is calculated

    A method for space-variant deblurring with application to adaptive optics imaging in astronomy

    Images from adaptive optics systems are generally affected by significant distortions of the point spread function (PSF) across the field of view, depending on the position of natural and artificial guide stars. Image reduction techniques circumventing or mitigating these effects are important tools to take full advantage of the scientific information encoded in AO images. The aim of this paper is to propose a method for the deblurring of the astronomical image, given a set of samples of the space-variant PSF. The method is based on a partitioning of the image domain into regions of isoplanatism and on applying suitable deconvolution methods with boundary effects correction to each region. The effectiveness of the boundary effects correction is proved. Moreover, the criterion for extending the disjoint sections to partially overlapping sections is validated. The method is applied to simulated images of a stellar system characterized by a spatially variable PSF. We obtain good photometric quality, and therefore good science quality, by performing aperture photometry on the deblurred images.     High Data Output and Automated 3D Correlative Light–Electron Microscopy Method

    Correlative light/electron microscopy (CLEM) allows the simultaneous observation of a given subcellular structure by fluorescence light microscopy (FLM) and electron microscopy. The use of this approach is becoming increasingly frequent in cell biology. In this study, we report on a new high data output CLEM method based on the use of cryosections. We successfully applied the method to analyze the structure of rough and smooth Russell bodies used as model systems. The major advantages of our method are (i) the possibility to correlate several hundreds of events at the same time, (ii) the possibility to perform three-dimensional (3D) correlation, (iii) the possibility to immunolabel both endogenous and recombinantly expressed proteins at the same time and (iv) the possibility to combine the high data analysis capability of FLM with the high precision–accuracy of transmission electron microscopy in a CLEM hybrid morphometry analysis. We have identified and optimized critical steps in sample preparation, defined routines for sample analysis and retracing of regions of interest, developed software for semi/fully automatic 3D reconstruction and defined preliminary conditions for an hybrid light/electron microscopy morphometry approach

    Commissioning and improvements of the instrumentation and launch of the scientific exploitation of OARPAF, the Regional Astronomical Observatory of the Antola Park

    The OARPAF telescope is an 80-cm-diameter optical telescope installed in the Antola Mount Regional Reserve, in Northern Italy. We present the results of the characterization of the site, as well as developments and interventions that have been implemented, with the goal of exploiting the facility for scientific and educational purposes. During the characterization of the site, an average background brightness of 22.40mAB (B filter) to 21.14mAB (I) per arcsecond squared, and a 1.5″ to 3.0″ seeing, have been measured. An estimate of the magnitude zero points for photometry is also reported. The material under commissioning includes three CCD detectors for which we provide the linearity range, gain, and dark current; a 31-orders échelle spectrograph with R ∼ 8500 to 15,000 and a dispersion of n = 1.39 × 10 − 6 px − 1λ + 1.45 × 10 − 4 nm / px, where λ is expressed in nm. The scientific and outreach potential of the facility is proven in different science cases, such as exoplanetary transits and active galactic nuclei variability. The determination of time delays of gravitationally lensed quasars, the microlensing phenomenon, and the tracking and the study of asteroids are also discussed as prospective science cases

    The application of Poisson data inversions is important in both specific and very different domains of applied sciences, such as microscopy, medical imaging and astronomy. The purpose of the book is to provide a comprehensive account of theoretical results, methods and algorithms related to the problem of image reconstruction from Poisson data in the framework of the maximum likelihood approach introduced by Shepp and Vardi. This is achieved by first discussing the application domains where this approach is important and their mathematical modelling, including the statistical properties of the data. The authors introduce the maximum likelihood approach which naturally arises from this modelling. Finally, a suitable variational formulation of this approach opens the door to all subsequent advancements and results. The description of the methods proposed in the literature is organized into a few large classes, discussing first the methods for differentiable regularization and subsequently those, more complex, for non-differentiable regularization

We restrict the presentation to the case of data affected by Poisson noise and of nonnegative solutions; both maximum likelihood and Bayesian approaches are considered. Numerical results are presented for discussing the practical behaviour of the SGP methods
